خلاصه يا چکيده : ‭7 cells in the presence of a low concentration of AgNPs. Method: The designed protocols were applied through five steps on the cells at different groups. At the first, cytotoxicity of AgNPs was determined. Then, the parameters of electroporation were optimized. At this stage the cells received 4 pulses with different strengths and durations at 1Hz frequency. In the third step, electric current with a frequency of 300 kHz was applied. In the fourth stage, different dual combinations of the treatments were investigated, and finally the synergism of the combinational treatments was evaluated. The cells viability was estimated by MTT assay after different treatments. Results and discussion: The data showed a synergism effect in the presence of AgNPs and electroporation with a 700 V/cm strength and 100, 300 and 500 microsecond durations. The greatest synergism was related to the duration of 100 microseconds. Also, in the voltages of 3, 6, and 9 Volt at the 300 kHz frequency, the synergism effects was recorded in the presence of AgNPs. In the presence of AgNPs, a greater synergism was determined by the triplet combinational treatment in comparison with the dual treatments. Conclusion: A significant cell death is provided after applying a combination therapy via a low dose of AgNPs, electrical pulses and low voltages of 300 kHz. In this study, electroporation and a low voltage at intermediate frequency in combination together are utilized in order to induce cytotoxicity on the MCF-Background: Considering some reports on the subject of significant antimicrobial properties of silver nanoparticles (AgNPs), the main aim of this study was designed on the base of utilizing the nanoparticles' toxicity to destroy cancer cells. Some studies have proved that AgNPs provide toxicity in the vital organs. Hence, in order to the safe clinical use of them, low dose administration is recommended but a favorable outcome will be obtained when the decreased dose is compensated by the other techniques. As alive cells consist of ions, polar molecules, and charged membranes, they response to electric fields. Electroporation is an electrical manner for pore creation in the cell membrane and transiently increases cell permeability. Therefore, some molecules which cell membrane is not naturally permeable to them can be transferred into the cells. In this technique, application of high strength and low duration electrical pulses with chemotherapeutic agents can be efficient in malignancies treatment.
