خلاصه يا چکيده : ‭Butylhydroperoxide, and 2 hours later 10 and 50 -Butylhydroperoxide with a concentration of 0.1mmol per kg was administered intraperitoneally. Groups III and IV received the same dose of tert-inflammatory and antioxidant properties. In order to more investigate the antioxidant properties of ghrelin this study was designed to evaluate the effect of ghrelin onoxidative stress in both in in vivo and in vitro models.Method: For in vivo study, 36 male rats were selected and divided into six groups of six.For the first group (the control group), a normal saline solution was injected. In group II, tert-Background: Ghrelin was identified by Kojima Masayasu and colleagues in 1999. This hormone is a peptide of 28 amino acids, secreted from the gastrointestinal tract, particularly the stomach during fasting. It has an important role in modulating energy, as well as anti‬إ‭g/kg of ghrelin was injected, resepectively.Groups V and VI received a single dose of 10 or 50‬إ‭g/kg , respectively.Twenty four hours after injection of the above materials, 8 to 10 ml blood sample was taken from the rats hearts in accordance with ethical standards. Blood samples were centrifuged for 10 min at 4‬░‭C with 3000 g. Serum was isolated according to the protocol for the measurement of biochemical markers of total antioxidant capacity (TAC), protein carbonyl and nitric oxide (NO).For in vitro study, the effect of ghrelin on HepG2 cell line was investigated. Cells in DMEM containing FBS 10 and 100 units / ml penicillin and 100 mg/ ml Streptomycin were treated with 10, 25 or 50 nmol ghrelin prepared in sterile PBS. After 30 min incubation at 37‬ ░‭Butylhydroperoxide was added with either 100 or 200 M, and after 24 hours of incubation, the percentage of living cells was examined by resazurin.Results: Ghrelin could diminish oxidative stress, proven through the reduction of protein carbonyls. Ghrelin had no significant effect on serum total antioxidant capacity. Ghrelin increased the serum level of nitric oxide as well. Ghrelin did not show any significant antioxidant effect on HepG2 cell line in this study.Conclusion: According to our results, ghrelin had antioxidant activity which seems to be mediated through the formation of nitric oxide.
