خلاصه يا چکيده : ‭siRNA for silencing of Brg1 gene.Results Our results showed for the first time that SMARCA4 (Brg1) expression was increased in KYSE30 cell line (ESCC cell line) compared with normal Esophageal tissue of ESCC patients and our previous study showed that MMPs enzymes were over expressed in ESCC patients (unpublished data). Also our in silico studies represent Brg1 and MMP2 interaction. Also silencing of SMARCA4(Brg1) leads to a reduction in MMP2 expression.Conclusions Our data indicated that chromatin remodeling machinery is a novel aspects in ESCC tumor biology and over expression of SMARCA4(Brg1) as an important member of SWI/SNF might be involved in the migration and invasion of ESCC tumoral cells.-PCR we evaluated SMARCA4(Brg1) expression in KYSE30 and normal esophageal tissue of ESCC patients. KYSE30 Cells were cultured in RPMI1640 supplemented with 10 fetal bovine serum. KYSE30 cells were transfected with Brg1-Introduction Esophageal Squamous Cell Carcinoma (ESCC) is the ninth common malignancy and the sixth most frequent cause of cancer mortality in Iran. Matrix metalloproteinases (MMPs) are known as important player enzymes during metastasis and invasion. Epigenetic regulation of gene expression can be carried out through chromatin remodeling enzymes such as SWI/SNF. Brg1also known as SMARCA4, is a catalytic subunit of SWI/SNF which is necessary for genes expression. Aberrant epigenetic modification in chromatin state should be precisely clarified to reveal probable unknown pathways of carcinogenesis and may introduce a novel targeted for therapeutic approaches.Purpose The aim of this study is elucidating the role of Brg1 in ESCC malignancy and to determine the effect of Brg1 on progression and migration based on our previous studies on genes expression.Materials and Methods Using Real Time‬
