خلاصه يا چکيده : ‭healing BALB/c mice model of Leishmania major infection in comparision to Amphothericin B, Meglumin Antimoniate and Placebo by measuring lesions size 3 times weekly for 7 weeks .We also used Limiting dilution assay to determine spleen and foot pad parasite burden of treated mice 21 and 48 days after infection. Results: Nimodipine was highly active against promastigote and intracellular amastigotes of leishmania major with ED50 values of 43 M and 23g. Although Early treatment with Nimodipine 1mg/kg lead to increased lesion size compared to control group, early treatment with higher doses and treatment of established lesions decreased lesions size comparable to Amphothericin B and Meglumin antimoniate. early treatment with combination of Nimodipine and Meglumin antimoniate resulted in lower spleen and foot pad parasite burden at day 21 compared to control group.Conclusion:Nimodipine has promising effects against promastigotes and amastigotes of leishmania major and in BALB/c mouse model of the disease.Further works are required to evaluate the effect of this drug in the course of leishmaniasis in human subjects.-Background: Cutaneous leishmaniasis is a common parasitic disease in many parts of the world. Pentavalent antimonials are the standard treatment in endemic regions but treatment failures are common and resistance is emerging.There is increasing evidence that calcium channel blockers such as Nimodipine may have promising antimicrobial and antiprotozoal effects. In the present study we evaluated the effect of calcium channel blocker nimodipine on the in vitro viability of promastigotes and amastigotes of leishmania major.We also evaluated the effect of Nimodipine on the course of leishmaniasis in BALB/c mouse model of disease .Method: We have used colorimetric alamar blue assay and a macrophage model to determine the effect of drug on promastigotes and amastigotes respectively.
