خلاصه يا چکيده : ‭time PCR we found that the CCR6 mRNA level was significantly increased in HAM/TSP patients and asymptomatic carriers compared to healthy group. Differences in mRNA levels between healthy and HAM/TSP group was (p<0.005) and differences between healthy and asymptomatic carriers was (p<0.041). Also the CXCR3 mRNA level was significantly decreased in asymptomatic carriers and HAM/TSP patients compared to healthy group (p<0.001). CXCL9 level increased significantly in HAM/TSP patients compared with asymptomatic carriers and healthy group however CXCL10 level was not significantly changed among three groups. Discussion: These data showed that chemokine receptors express in different conditions and times in inflammatory diseases. As far as the patients in this study were in the early phase of the HAM/TSP disease it seems that increased in the CCR6 expression in the peripheral blood play a pivotal role in inflammation while the expression of CXCR3 expressing cells frequency doesnt change in this time of the disease.-1 carriers. Results: According to data obtained from Real-I Proviral load was measured in patients with HAM / TSP and HTLV-Time PCR technique was performed. To evaluate the amount of CXCL9 and CXCL10 in the serum, ELISA technique was carried out on sera of all subjects. HTLV-1 carriers (22 subjects) and healthy people (20 subjects) were included and blood samples (10 ml) were obtained from each subject. To synthesize cDNA, peripheral blood lymphocytes were separated with centrifugation density gradient and RNA was extracted and then cDNA synthesized. In order to quantify gene expression of CXCR3 and CCR6 chemokine receptors, Taqman probes and primers were designed and Real-I is an oncogenic human retrovirus that in the 5 10 of infected individuals causes one of two types of disease: ATL and HAM/TSP. Following infection with the virus changes in the lymphocyte chemokine and chemokine receptors gene expression is happened which confers the ability to T cells to migrate to CNS. Increased expression of CCR6 chemokine receptor on Th17 cells cause these cells transmigrate across blood brain barrier and enter CNS. On the other hand increased expression of CXCR3 on the Th1 and CTLs intensified the CNS inflammation.
